Yes, I have installed MPE and it was working normally but I had this problem since 1 or 2 months : perhaps when I have updated Thunderbird (but it was before thunderbird 3).
I decided to ask for this problem since the extension Lightning works with Thunderbird 3.

For FJ,
I had not much time to reply this afternoon.
Now I can precise my first answer : I would say that the folder "C:\Documents and Settings\" exists under XP and not in Vista.

No more problem, after I change the database repertory as you told me, !!!
I learn a lot : I ldidn't know the error console in Thunderbird.

Under Windows Vista, MPE 1.7.4 and with a Sony Ericsson w660i, I obtain an error message (in French) that tells it's impossible to synchronize the adress contacts or the agenda of Thunderbird 3 / Lightning 1.0b1.
The same happened with the old mpe addin.
Can you open the "Advanced"-Dialog for ...
